  1. The vitamin E family comprised of eight chemically distinct compounds: 4 tocopherols and 4 tocotrienols (alpha, beta, gamma and delta). Tocotrienols differ from tocopherols by having an unsaturated side tail that results in significantly different biological activities.

    How does palm tocotrienol complex confer neuroprotection?
    1) Tocotrienol has been reported to cross the blood-brain barrier. Alpha-tocotrienol was reported in the Journal of Biological Chemistry to be much more potent than the regular alpha-tocopherol in protecting the brain cells / neurons from glutamate-induced toxicity. For full protection, nanomolar level of tocotrienol was required whereas the regular tocopherol requires micromolar level – a 1000X more potent. Carotech’s research collaboration with the Ohio State University Medical Center on Tocomin® and potent neuro-protection has recently received a research grant in excess of USD 1 million from the United States’ National Institute of Health.

    2) Palm tocotrienol complex is a much more potent antioxidant than tocopherol. It has been shown by published research paper that alpha-tocotrienol is 40 – 60 times more potent than alpha-tocopherol in the prevention of lipid peroxidation.

    3) Tocotrienols, with their unsaturated side chain, allow them to pass through the membrane bi-layer (mainly made up of unsaturated fatty acid) in a more efficient manner and faster rate compared to the all saturated tocopherol.

    In summary, there are 2 main actions how palm tocotrienol complex confer neuroprotection:

    a. through powerful antioxidant function as mentioned above

    b. through mechanisms other than antioxidant function - Tocotrienol protects neuron death by other mechanism unrelated to its antioxidant activity. In a number of NIH-sponsored studies, Researchers at the Ohio State Medical Center showed that alpha-tocotrienol, but not alpha-tocopherol, prevented neuron’s death at extremely low concentration (nanomolar, 10-9) by regulating specific mediators of cell death (Sen CK et al., 2000). Tocotrienol-treated neurons maintained healthy growth and motility even in the presence of excess neurotoxic agent. The neuroprotective property seen at such low concentration of tocotrienol is independent of its antioxidant activity, as tocotrienols do not exhibit antioxidant properties at nanomolar concentrations. Tocotrienols only begins to show its antioxidant effects at micromolar concentrations (Khanna et al., 2006).

    The same researchers went on to demonstrate that oral supplementation of tocotrienols (Tocomin®) reaches rats’ brain in concentration that protected against stroke. There is reduced volume of cerebral infarct in tocotrienol-supplemented rats compared with un-supplemented controls (Khanna S et al., 2005).

    A further study showed that Tocomin®SupraBio™ supplementation in healthy women achieved plasma levels of tocotrienols 12 to 30 times more than the concentration of α-tocotrienol required to completely prevent stroke-related neurodegeneration (Khosla P et al., 2006).

    With the above research, a clinical trial is now ongoing to study the neuroprotective effects of palm tocotrienol complex. This is the first study of its kind and the world’s largest clinical trial utilizing MRI brain to evaluate the efficacy of palm tocotrienol complex in neuroprotection and antiatherogenic function in human. The results of this clinical trial will shed more light on the benefits of palm tocotrienol complex (Tocomin®) in both acute neurological disorders (e.g. stroke) and chronic neurodegenerative disorders (e.g. Alzheimer’s disease; Parkinson’s disease).
